《美國隊長》系列形象誕生於上世紀40年代，故事背景設定在二戰時期，主角斯蒂夫·羅傑斯（Steve Rogers）作為超級士兵實驗的成功受體化身“美國上尉”，對抗恐怖的納粹組織。本次的游戲將根據即將於2011年7 月上映的同名影片改編而成，美國上尉為了阻止納粹恐怖行動特殊組織Hydra 中的反派紅骷髅與邪惡科學家Arnim Zola向聯合國發射飛彈的陰謀，單槍匹馬潛入敵方要塞展開冒險。
